Combination Therapy (CagriSema): Cagrilintide has been co-formulated with semaglutide, a GLP-1 receptor agonist, resulting in the combination therapy known as CagriSema. In a 32-week phase 2 trial involving participants with type 2 diabetes, CagriSema treatment resulted in a mean HbA1c reduction of 2.2 percentage points and a mean body weight reduction of 15.6%. These results were superior to those observed with semaglutide or cagrilintide alone.

Phase 3 REDEFINE 1 Trial: In a 68-week phase 3 trial, participants receiving CagriSema experienced a 22.7% reduction in body weight, compared to 11.8% with cagrilintide alone and 16.1% with semaglutide alone. While these results fell slightly short of the anticipated 25% weight loss, they still represent a substantial reduction in body weight.

Cagrilintide, both as monotherapy and in combination with semaglutide, shows promise in managing obesity and improving metabolic health. Ongoing research aims to further elucidate its therapeutic potential and long-term safety profile.
